Puyo Puyo Fever 2 (ぷよぷよフィーバーチュー！) is the sequel to Puyo Puyo Fever. As with the other games in the Fever series, it takes place in Primp Town, home to the magic school of the last game, now used for the game's 9 courses. It also contains a tower (for endless puyo modes), a shop (to buy items, run by Oshare Bones, however Rider is seen there as well), a town hall (to change options and save), a museum (for viewing unlocked music, videos, etc.), and a playground (for multiplayer). You can explore these areas and talk to the people there, where they may give you items.
The courses in this game consist of multiple encounters the character makes until they reach their objective. Each encounter begins with a short conversation, always leading to a battle, excluding the final sequence of dialog in each course. There is now a new items system which affects the gameplay and beginning of battles. Some items allow you to play as another character. The majority do helpful things, like dumping nuisance puyo on an opponent at the beginning, making attacks more powerful, setting up a chain for you to start with, or starting you with a couple points on your fever counter. Common items can be bought at Oshare Bones' shop.
The game boasts nine courses, three times that of the original Puyo Puyo Fever. Each of the three main characters (Amitie, Raffine, and Sig) has three courses, each with a different story.
Three save files are allowed, as if to track progress for multiple players.
A pink variation of the nuisance puyo, known as CHU Puyo, were introduced in this game and do not appear in any other game to date. However these only appear in Endless CHU Panic mode where ordinary trash does not, thus have no effect on gameplay and are a purely graphical modification.

Courses

RunRun
The RunRun (るんるん, runrun) course in Puyo Puyo Fever 2 is similar to the original RunRun course in that it's designed to get the player started. It starts at a dialog with accord. Each player face three others: 2 classmates and then a final test by Accord.

The WakuWaku (わくわく, wakuwaku) course is also similar to the original course in a similar manor.
Raffine
This course begins with Rider beginning to talk, when suddenly, Butler rushes to Raffine and Rider is literally plowed across to another location. After a short dialog with Butler, Rider returns, dizzy, and the course begins.
The final dailog shows Lemres turn Ocean Prince into his human form, Salde, as Butler tries to catch up with him, who struggles away from Lemres and Raffine. Raffine then looks at the item she gets, the Shellfish of thinking, in disgust. The screen fades and the credits roll.
Amitie
As the course begins, Amitie is found talking to Onion Pixy.
As the course ends, Amitie seems to have been embarassed and made a mistake. Accord appears and says something, the screen fades out and the credits roll.
Sig
Sig's course begins on a dialog with Accord. He then meets with Onion Pixy, where Onion Pixy Girl falls in love with Sig, and Onion Pixy fights him. After this, he meets with Rider, where Rider inqueries about his hair. This somehow lands them into a match.
This course ends with Sig reading something that appears to be a message from or to Lemres. Then Sig lifts and drops throws Lemres against the ground for an unknown reason.
HaraHara
The HaraHara (はらはら, harahara) course is similar as well.
Raffine
The course begins with Lemres talking to Raffine. They seem to be trying to figure something out. Raffine seems shocked at some of the things said. Raffine then meets with Klug. Klug seems to be rather meniachal preceding their match.
